Composition per liter:

Glucose ......................................................................................... 2.0g

Pancreatic digest of casein............................................................ 1.0g

Yeast extract.................................................................................. 0.5g

L-Cysteine·HCl............................................................................ 0.05g

Resazurin .................................................................................. 0.001g

Seawater.................................................................................750.0mL

Tris·HCl buffer

(0.2M solution, pH 7.5) ...................................................250.0mL

pH 7.5 ± 0.2 at 25°C

Preparation of Medium: Prepare and dispense medium under

100% N2. Combine components. Mix thoroughly. Sparge with 100%

N2. Adjust pH to 7.5 while continuing to sparge with 100% N2. Anaer

obically distribute into tubes or flasks. Autoclave for 15 min at 15 psi

pressure–121°C.

Use: For the cultivation and maintenance of Spirochaeta isovalerica.
