Composition per 1001.2mL:

Disodium-ß-glycerophosphate............................9.5g

Pancreatic digest of casein..................................5.0g

Meat peptone.......................................................5.0g

Papaic digest of soybean.....................................5.0g

Yeast extract........................................................2.5g

Ascorbic acid ......................................................0.5g

MgSO4·7H2O ....................................................0.25g

Lactose solution ............................................ 50.0mL

CaCl2 solution................................................. 1.2mL

pH 7.15 ± 0.05 at 25°C

Lactose Solution:

Composition per 50.0mL:

Lactose................................................................8.0g

Preparation of Lactose Solution: Add lactose

to distilled/deionized water and bring volume to

50.0mL. Mix thoroughly. Filter sterilize.

CaCl2 Solution:

Composition per 100.0mL:

CaCl2·2H2O.......................................................14.7g

Preparation of CaCl2 Solution: Add 14.7g of

CaCl2·2H2O to distilled/deionized water and bring

volume to 100.0mL. Mix thoroughly. Filter sterilize.

Preparation of Medium: Add components, ex

cept lactose solution and CaCl2 solution, to distilled/

deionized water and bring volume to 950.0mL. Mix

thoroughly. Autoclave for 15 min at 15 psi pressure–

121°C. Aseptically add 50.0mL of sterile lactose so

lution and 1.2mL of sterile CaCl2 solution. Mix thor

oughly. Aseptically adjust pH to 7.15 ± 0.05.

Aseptically distribute into sterile tubes or flasks.

Use: For the cultivation of Streptococcus thermophi

lus.
