Mineral Salts with Butane

Composition per liter of tap water:

(NH 4 ) 2 HPO 4 ..................................................................................8.0g
Na 2 HPO 4 ·12H 2 O...........................................................................2.5g
KH 2 PO 4 .........................................................................................2.0g
MgSO 4 ·7H 2 O................................................................................0.5g
Yeast extract...........................................................................100.0mg
CaCl 2 ·2H 2 O .............................................................................60.0mg
FeSO 4 ·7H 2 O.............................................................................30.0mg
MnCl 2 ·4H 2 O .............................................................................60.0μg
CuSO 4 ·5H 2 O.............................................................................15.0μg
pH 7.1 ± 0.2 at 25°C
Preparation of Medium: Add components to distilled/deionized
water and bring volume to 1.0L. Mix thoroughly. Distribute into tubes
or flasks. Autoclave for 15 min at 15 psi pressure–121°C. Incubate in-
oculated medium in 88% air + 7% n-butane + 5% CO 2 .
Use:  For the cultivation and maintenance of Pseudomonas butano-
vora.
