Composition per liter:

ACES buffer (2-[(2-amino-2-oxoethyl)-
amino]-ethane sulfonic acid)................................10.0g
Charcoal, activated .........................................2.0g
α-Ketoglutarate.............................................1.0g
Bovine serum albumin solution..............................10.0mL
L -Cysteine·HCl·H2O solution.............................10.0mL
Fe4(P2O7)3 ·9H2O solution.................................10.0mL
pH 6.9 ± 0.2 at 25°C
Bovine Serum Albumin Solution:
Composition per 10.0mL:
Bovine serum albumin.........................................0.1g
Preparation of Bovine Serum Albumin Solution: Add bovine
serum albumin to distilled/deionized water and bring volume to
10.0mL. Mix thoroughly. Filter sterilize.
L -Cysteine·HCl·H2O Solution:
Composition per 10.0mL:
L- Cysteine·HCl·H2O.......................................0.4g
Preparation of L -Cysteine·HCl·H2O Solution: Add L -
cysteine·HCl·H2O to distilled/deionized water and bring volume to
10.0mL. Mix thoroughly. Filter sterilize.
Fe4(P2O7)3 ·9H2O Solution:
Composition per 10.0mL:
Fe4(P2O7)3 ·9H2O........................................0.25g
Preparation of Fe4(P2O7)3 ·9H2O Solution: Add Fe4(P2O7)3 ·9H2O
to distilled/deionized water and bring volume to 10.0mL. Mix thoroughly.
Filter sterilize.
Preparation of Medium: Add components—except Fe4(P2O7)3 ·9H2O
solution, L -cysteine·HCl·H2O solution, and bovine serum albumin solu-
tion—to distilled/deionized water and bring volume to 970.0mL. Mix
thoroughly. Adjust medium to pH 6.9 with 1N KOH. Heat gently and bring
to boiling for 1 min. Autoclave for 15 min at 15 psi pressure–121°C. Cool
to 50°–55°C. Aseptically add 10.0mL of sterile bovine serum albumin so-
lution, the Fe4(P2O7)3 ·9H2O solution, and the L -cysteine·HCl·H2O solu-
tion. Mix thoroughly. Pour into sterile Petri dishes with constant agitation
to keep charcoal in suspension.
Use: For the isolation, cultivation, and maintenance of Legionella
pneumophila and other Legionella species from environmental and
clinical specimens.
