Composition per liter:

Pancreatic digest of casein................................20.0g

Lactose................................................................5.0g

NaCl....................................................................5.0g

K2HPO4.............................................................2.75g

KH2PO4.............................................................2.75g

Sodium lauryl sulfate..........................................0.1g

pH 6.8 ± 0.2 at 25°C

Source: This medium is available as a premixed

powder from BD Diagnostic Systems.

Preparation of Medium: Add components to

distilled/deionized water and bring volume to 1.0L.

Mix thoroughly. Distribute into tubes containing an

inverted Durham tube in 10.0mL volumes. Auto

clave for 12 min at 15 psi pressure–121°C. Cool

broth quickly to 25°C. For testing water samples with

10.0mL volumes, prepare medium double strength.

Use: For the detection of coliform bacteria, including

for the enumeration of coliform bacteria by the multi

ple-tube fermentation technique.
