Sporulation Broth

Composition per liter:

Polypeptone™............................................................................. 15.0g

Na2HPO4..................................................................................... 11.0g

Starch, soluble............................................................................... 3.0g

Yeast extract.................................................................................. 3.0g

Sodium thioglycolate .................................................................... 1.0g

MgSO4, anhydrous........................................................................ 0.1g

pH 7.8 ± 0.1 at 25°C

Preparation of Medium: Add components to distilled/deionized

water and bring volume to 1.0L. Mix thoroughly. Distribute into tubes

in 15.0mL volumes. Autoclave for 15 min at 15 psi pressure–121°C.

Use: For the cultivation and observation of sporulation of Clostridium

perfringens.
