Artificial sea water, concentrated...........................................190.0mL
pH 7.2 ± 0.2 at 25°C
Artificial Sea Water, Concentrated:
Composition per 1.0L:
NaCl............................................................................................99.4g
MgSO 4 ·7H 2 O............................................................................23.76g
MgCl 2 ·6H 2 O..............................................................................18.12g
CaCl 2 ·2H 2 O...................................................................................5.2g
KCl..............................................................................................2.56g
Preparation of Artificial Sea Water, Concentrated: Add components to distilled/deionized water and bring volume to 1.0L. Mix thoroughly. Adjust pH to 7.1–7.3. Autoclave for 15 min at 15 psi pressure–121°C. Cool to room temperature.
Preparation of Medium: Add components, except concentrated artificial sea water, to distilled/deionized water and bring volume to 810.0mL. Mix thoroughly. Adjust pH to 7.1–7.3. Autoclave for 15 min at 15 psi pressure–121°C. Cool to 60°C. Add 190.0mL concentrated artificial sea water. Mix thoroughly. Aseptically distribute into tubes or flasks.
Use:  For the cultivation of Alcanivorax jadensis.
