Polypectate Gel Medium

Composition per liter:

Sodium polypectate ..........................................70.0g

K2HPO4...............................................................5.0g

Peptone ...............................................................5.0g

KH2PO4...............................................................1.0g

CaCl2·2H2O ........................................................0.6g

pH 7.0 ± 0.2 at 25°C

Preparation of Medium: Add components, except

sodium polypectate, to 500.0mL of boiling water. Put

into a blender and mix on low speed to dissolve. Add the

sodium polypectate slowly with the blender on low

speed to minimize air bubbles. Adjust pH to 7.0. Bring

volume to 1.0L with distilled/deionized water. Auto

clave for 15 min at 15 psi pressure–121°C. Cool to 45°–

50°C. Pour into sterile Petri dishes.

Use: For the cultivation of microoganisms and de

tection of pectate-degrading enzymes.
