Composition per liter:

NaCl..................................................................65.0g

Glucose ...............................................................5.0g

Pancreatic digest of casein..................................5.0g

Yeast extract........................................................5.0g

NaN3 ...................................................................0.4g

Methylene Blue..............................................10.0mg

Penicillin...........................................................650U

pH 8.0 ± 0.2 at 25°C

Source: This medium is available as a premixed

powder from BD Diagnostic Systems.

Caution: Sodium azide is toxic. Azides also react

with metals and disposal must be highly diluted.

Preparation of Medium: Add components, ex

cept penicillin, to distilled/deionized water and bring

volume to 1.0L. Mix thoroughly. Gently heat and

bring to boiling. Autoclave for 15 min at 15 psi pres

sure–121°C. Cool to 25°C. Aseptically add penicil

lin. Mix thoroughly. Add sufficient amount of

Enterococci confirmatory broth to cover half the

slant of Enterococci confirmatory agar.

Use: For the identification of enterococci from water

by the confirmatory test.
